Profiling T cell interaction and activation through microfluidics-assisted serial encounter with APCs

Adoptive T cell therapy that collected from the patient’s blood for treating cancer and chronic infections has revolutionized field of oncology personalized medicine. However, such highly activated cells have specific receptor (TCR) against target disease are usually greatly outnumbered by not-activated due to stochastic generation TCR V(D)J recombination, approximately 106∼107 possible TCR. In this report, purpose representing lymph node where can migrate on surface antigen presenting (APCs) activation or sequentially interact multiple APCs, an open-type PDMS microfluidic device been developed. This mimics microenvironment a scan, contact with APC structure plan collect conduct downstream analysis later on. By measuring profiling Ca2+ flux interacted APC, threshold classifying whether is not was calculated. new tool expected be useful in providing insight strategy basic biology.
